Gene Expression Analysis
in situ hybridization- detection of spacial and temporal gene expression patterns (using RNA probes)
qPCR- technique used to monitor the amplification of specific genes in “real-time”
fluorescent “in vivo” monitoring of gene expression using stable transgenic lines
Standard Molecular Techniques
Although The Holowiecki lab uses many techniques that are specific to zebrafish, we also use techniques common to all molecular biology labs. For example, generation of the RNA probes required to perform in situ hybridization may entail multiple techniques prior to actual in situ analysis. These include: RNA isolation, cDNA synthesis, sub-cloning, restriction analysis and gel electrophoresis, primer design, DNA sequencing, and PCR.
